小分子(small molecules)是指分子量小于1000道爾頓(尤其小于400道爾頓),并具有生物學(xué)功能的化合物。此類化合物與細(xì)胞因子和蛋白不同,其可直接穿過細(xì)胞膜進(jìn)入細(xì)胞,進(jìn)而發(fā)揮相應(yīng)的功能,適用于各種屬的細(xì)胞。小分子化合物的作用靶點眾多,已廣泛應(yīng)用于干細(xì)胞,類器官,免疫學(xué),神經(jīng)生物學(xué),表觀遺傳學(xué),細(xì)胞凋亡,離子通道,腫瘤學(xué)和信號轉(zhuǎn)導(dǎo)等生命科學(xué)中諸多重要的研究領(lǐng)域。

❶ Gastrin I: 胃泌素是胃腸道一種非常重要的多肽類激素,主要由 G 細(xì)胞分泌。腸道及肝臟類器官培養(yǎng)時,需要添加胃泌素來延長類器官的存活時間。
❷ A83-01: Activin/NODAL/TGF-beta 通路抑制劑,其抑制 ALK5,ALK4 和 ALK7等激酶。一般用于肝臟、前列腺以及乳腺類器官的培養(yǎng)。
❸ Y27632: 是Rho相關(guān)絲氨酸-蘇氨酸蛋白激酶 (ROCK) 家族的小分子特異性抑制劑,可以阻止干細(xì)胞的凋亡,一般在種板培養(yǎng)的第一次加入,后續(xù)換液培養(yǎng)不必添加該小分子。
❹ SB202190: 是一種高效的 p38 MAPK 抑制劑,靶向作用于 p38α/β?捎糜谖改c道、乳腺類器官的培養(yǎng)。
❺ Nicotinamide: 是一種B3維生素,是NAD 和 NADP的活性成分,在細(xì)胞生理學(xué)中發(fā)揮重要作用,可用于胃腸道、肝臟以及乳腺類器官的培養(yǎng)。
❻ Forskolin: 可以激活腺苷環(huán)化酶,常用于提高細(xì)胞內(nèi) cAMP 的水平。Forskolin 誘導(dǎo)多種類型細(xì)胞的分化并激活孕烷X受體 (PXR) 和法尼醇X受體(FXR),可對心臟產(chǎn)生正性肌力作用,并具有血小板抗凝集和降壓作用。據(jù)報道,它與其他小分子聯(lián)合使用,可誘導(dǎo)成纖維細(xì)胞重編程為誘導(dǎo)多能性干細(xì)胞,在肝臟類器官培養(yǎng)的時候必須要添加。
❼ Prostaglandin E2: 前列腺素 E2,PGE2。PGE2調(diào)節(jié)許多生理系統(tǒng),包括消化系統(tǒng)和免疫系統(tǒng)等。它與特異的受體結(jié)合后可介導(dǎo)細(xì)胞增殖、分化,在肝臟類器官和前列腺類器官培養(yǎng)時需要添加。
❽ CHIR99021: CHIR99021是糖原合成酶激酶3β抑制劑,可誘導(dǎo)人胚胎干細(xì)胞向內(nèi)胚層分化,在腎臟以及視網(wǎng)膜類器官培養(yǎng)時需要。
❾ N-acetyl-L-Cysteine: N-乙酰半胱氨酸,它是一種 ROS 抑制劑,具有抗氧化作用,在大多數(shù)類器官的培養(yǎng)過程中需要添加。
