背景:
在真核生物中,染色質(zhì)結(jié)構(gòu)的調(diào)整在轉(zhuǎn)錄調(diào)控中起重要作用。核小體,由DNA纏繞在八個(gè)核心組蛋白上組成(H2A,H2B,H3和H4各兩個(gè))是染色質(zhì)的基本結(jié)構(gòu)單位。核心組蛋白的氨基末端進(jìn)行著各種翻譯后修飾,包括乙酰化,甲基化,磷酸化,泛素化。這些修改發(fā)生在各種刺激下,并直接影響染色質(zhì)與轉(zhuǎn)錄因子的可親近性,因此,影響基因表達(dá)。在大多數(shù)物種中,組蛋白H2B主要乙;奈稽c(diǎn)為L(zhǎng)ys5,12,15,20。組蛋白H3主要乙;稽c(diǎn)為L(zhǎng)YS9,14,18,23,27,和56。在一些生物中組蛋白H3的 Lys9乙;坪跻呀(jīng)在蛋白沉積和染色質(zhì)組裝中占據(jù)主導(dǎo)作用。組蛋白H3的Ser10,Ser28和Thr11磷酸化與有絲分裂和減數(shù)分裂過(guò)程中染色體凝集有緊密相關(guān)。組蛋白H3的THR3磷酸化在許多物種中是高度保守的并由激酶haspin催化。在哺乳動(dòng)物細(xì)胞中的免疫組織化學(xué)染色的磷酸化特異性抗體揭示了組氨酸H3的Thr3在有絲分裂前期的磷酸化和后期的去磷酸化。
