Aberrant miRNA Expression in Human Breast Oncogenesis

Abstract:
Breast cancer is believed to develop in a multistep process with premalignant lesions preceding invasive carcinoma. While a growing body of recent work has demonstrated the role of microRNAs in cancer, little is known about the role of microRNAs in precancerous events that precede malignant transformation.In this work we used Agilent miRNA microarray platform to characterize miRNA expression changes associated with breast oncogenesis using a multistep cell line model that mimics the progression of breast cancer, from benign to premalignant to malignant transformation, and with corresponding mouse xenografts. Additionally, we used a bioinformatics tool developed in our laboratory, miR-AT, to identify biological processes and pathways associated with transcripts targeted by the miRNAs.


Integrative Analysis of Genomic Imbalances in Cancer

Abstract:
Recurring non-random chromosome aberrations have been identified in hematological malignancies (leukemia and lymphoma) and soft tissue sarcomas. Among the various clonal aberrations, e.g. gene amplifications,deletions, inversions and duplicalions, chromosome lranslocations have been shown to have an important role in cancer development and they have been used as markers for diagnosis, prognosis and prediction of response to treatment . Current challenge is to understand the genetic complexity of solid tumors, because recurring chromosome abnormalities are difficult to ascertain in solid tumors by conventional karyotyping due to difficulties in chromosome preparation, poor morphology and high complexity of karyotype and clonal heterogeneity. High density oligonucleotide array-based comparative genomic hybridization (array CGH) is emerging as a powerful method for comprehensive analysis of unbalanced chromosome aberrations. In this presentation I will discuss the application of array CGH technology in solid tumor cancer research for the discovery of novel molecular markers.
