Roundabout homolog 1, also named ROBO1, Deleted in U twenty twenty and H-Robo-1, is a single-pass type I membrane protein which belongs to the neural adhesion molecule (NCAM) family of receptors. ROBO1 contains 3 fibronectin type-III domains and 5 Ig-like C2-type (immunoglobulin-like) domains. It is widely expressed, with exception of kidney. It is a receptor for SLIT1 and SLIT2 which are thought to act as a molecular guidance cue in cellular migration, including axonal navigation at the ventral midline of the neural tube and projection of axons to different regions during neuronal development. Defects in Roundabout homolog 1 may be a cause of breast and lung cancer. ROBO1 maps within a region of overlapping homozygous deletions characterized in both small cell lung cancer cell lines (SCLC) and in a breast cancer cell line.
