CD47, also known as Integrin Associated Protein(IAP) and OA3, is an integral membrane protein belonging to the immunoglobulin superfamily. Human CD47 contains an extracellular domain (ECD) with a single Ig-like domain, five membranespanning regions with short intervening loops, and a C terminal cytoplasmic tail. Within the ECD, human CD47 shares 63% sequence identity with mouse and rat CD47. The ubiquitously expressed CD47 binds to SIRP family members on macrophages, neutrophils, and T cells. CD47 interacts with SIRPA as a receptor, which prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells. CD47 interacts with SIRPG, which mediates cell-cell adhesion, enhances superantigen-dependent T-cell-mediated proliferation and costimulates T-cell activation. It has been shown that CD47 associates in cis with Fas on T cells and enhances Fas-mediated apoptosis. CD47 plays important roles in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. CD47 also functions in memory formation and synaptic plasticity in the hippocampus.

·Description
Recombinant Human CD47 produced by transiently transfected human cell is the extracellular domain (Accession#Q08722)( Gln19 – Pro139 ) of human CD47 fused with a polyhistidine tag at the C-terminus.

·Quality Control

Biological activity:
The ED50 of recombinant human CD47 is 0.5-2.0μg/mL as measured by its ability to antagonize red blood cell adhesion to immobilized rhSIRPα.

Purity: Greater than 90.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Amino-Acid Sequence: Predicted N terminal amino acid is Gln19.
Endotoxin:Less than 0.1ng/μg (1EU/μg) of rHu CD47.

 ·Formulation
Sterile and Lyophilized from a 0.2 μm filtered solution in PBS.

·Reconstitution
It is recommended to reconstitute the lyophilized rHu CD47 in sterile water not less than 100μg/ml.

·Storage
Lyophilized rHuCD47 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHuCD47 should be stored at 4°C between 2-7 days and aliquots of reconstituted samples containing 0.1% BSA are stable up to 3 months below -18°C for future use.
