CD47, also known as Integrin Associated Protein(IAP) and OA3, is an integral membrane protein belonging to the immunoglobulin superfamily. Human CD47 contains an extracellular domain (ECD) with a single Ig-like domain, five membranespanning regions with short intervening loops, and a C terminal cytoplasmic tail. Within the ECD, human CD47 shares 63% sequence identity with mouse and rat CD47. The ubiquitously expressed CD47 binds to SIRP family members on macrophages, neutrophils, and T cells. CD47 interacts with SIRPA as a receptor, which prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells. CD47 interacts with SIRPG, which mediates cell-cell adhesion, enhances superantigen-dependent T-cell-mediated proliferation and costimulates T-cell activation. It has been shown that CD47 associates in cis with Fas on T cells and enhances Fas-mediated apoptosis. CD47 plays important roles in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. CD47 also functions in memory formation and synaptic plasticity in the hippocampus.
