CD319, also known as CD2 like receptor-activating cytotoxic cells (CRACC), SLAM family member 7(SLAMF7)and CS1, is a single-pass type I membrane protein and a member of the CD2 family of cell surface receptors.CD319 is expressed in NK cells, activated B-cells, NK-cell line but not in promyelocytic, B-, or T-cell lines. The extracellular region of CD319 contains one Ig-like C2-type domain. CD319 mediates NK cell activation through ERK-mediated pathway dependent on the adaptor EAT-2 but not the related adaptor SAP. The cytoplasmic domain of CD319 contains immunoreceptor tyrosine-based switch motifs (ITSM),which upon phosphorylation, can bind to SAP. In the absence of EAT-2, CD319 exerts inhibitory functions.
·Description
Recombinant Human SLAMF7/CD319 produced by transiently transfected human cell is the extracellular domain (Accession # Q9NQ25) ( Ser23 – Met226 ) of human SLAMF7/CD319 fused with a polyhistidine tag at the C-terminus.
·Quality Control
Purity: Greater than 90.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Amino-Acid Sequence: Predicted N terminal amino acid is Ser23.
Endotoxin: Less than 0.1ng/μg (1EU/μg) of rHuCD319.
·Formulation
Sterile and Lyophilized from a 0.2 μm filtered solution in PBS.
·Reconstitution
It is recommended to reconstitute the lyophilized recombinant Human CD319 in sterile water at no less than 100 μg/ml.
·Storage
Lyophilized rHuCD319 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHuCD319 should be stored at 4°C between 2-7 days and aliquots of reconstituted samples containing 0.1% BSA are stable up to 3 months below -18°C for future use.
Please avoid freeze-thaw cycles.
