Human interleukin 7 (IL-7) is a potent lymphoid cell growth factor stimulating the proliferation of lymphoid progenitors. IL7 can associate with the hepatocyte growth factor (HGF) to form a hybrid cytokine that functions as a pre-pro-B cell growth-stimulating factor. Human IL7 cDNA encodes a 177 amino acid precursor protein containing a 25 aa signal peptide and a 152 aa mature protein. Human and mouse IL7 share 65% sequence identity in the mature region and both exhibit cross-species activity.
IL-7 signals via IL-7 receptor(IL7R) activating multiple pathways including JaK/STAT,PI3K/AKT pathways,which regulate lymphocyte survival, glucose uptake, proliferation and differentiation. IL-7 is also associated with cytoplasmic IL2-R gama for signal transduction.

Recombinant Human IL-7 produced in E. coli is a single, non-glycosylated, polypeptide chain containing 152 amino acids and having a molecular mass of 17.4kDa.
·Quality Control
Biological activity: The ED50 of rHuIL7 as determined by the dose dependent stimulation of mouse IxN/2b cell proliferation was less than 0.5 ng/ml, corresponding to a specific activity of 2.0 x 106 IU/mg.
Purity: Greater than 95.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Amino-Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Asp-Cys-Asp-Leu-Glu.
Endotoxin: Less than 0.1ng/μg (IEU/μg) of rHuIL-7 as determined by LAL test.
·Formulation
Sterile and Lyophilized aliquots with a 0.2um filtered protein solution containing 20mM PB pH8.0,300mM NaCl.
·Reconstitution
It is recommended to reconstitute the lyophilized rHuIL-7 in sterile 18MΩ-cm H2O not less than 100μg/ml, which can then be further diluted to working solutions.
·Storage
Lyophilized rHuIL-7 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHuIL-7 should be stored at 4°C between 2-7 days and aliquots of reconstituted samples are stable up to 3 months below -18°C for future use.
Please avoid freeze-thaw cycles.
